Anatomopathological study in BALB/c mice brains experimentally infected with Toxoplasma gondii BJID
Silva,Marcos Gontijo da; Lino Junior,Ruy de Souza; Costa,Tatiane Luiza da; Soares,Joanna D´arc Herzog; Amaral,Waldemar Naves do; Avelino,Mariza Martins; Castro,Ana Maria de.
Toxoplasmosis is one of the most important diseases of the nervous central system, leading to severe symptoms and, many times, irreversible sequelae. This work demonstrated the main anatomopathological lesions caused by Toxoplasma gondii in brains from experimentally infected BALB/c mice. We analyzed 51 cases of mice that developed toxoplasmosis after experimental infection by intraperitoneal inoculation of blood, amniotic liquid and cerebrospinal fluid from fetuses, newly born children and pregnant women with clinical and laboratory signals of toxoplasmosis. In all experiments where we detected the parasite in mice we also detected pathological lesions in the animal brains with great polymorphism between experiments. Edema was the most found lesion in all...

Cytomegalovirus (CMV) disease is a major cause of morbidity and mortality in solid organ transplantation. Disseminated toxoplasmosis after liver transplantation is a rare but fatal event. Serologic screening of the donor and the recipient is essential to prophylactic management, early diagnosis and therapeutic strategies to minimize the consequences of these infections. The aim of the present study was to determine the seroprevalence of CMV and Toxoplasma gondii (TG) in a Brazilian liver transplant waiting list (LTWL). Serological data were collected from 44 candidates on the LTWL between May 2003 and November 2004. Serological investigation of antibodies IgM and IgG against CMV (anti-CMV) and TG (anti-T. gondii) was performed using fluorometry commercial...

Detection of Toxoplasma gondii DNA in sera samples of mice experimentally infected J. Venom. Anim. Toxins incl. Trop. Dis.
Langoni,H.; Das Dores,C. B.; Silva,R. C.; Pezerico,S. B.; Castro,A. P. B.; Da Silva,A. V..
Detection of Toxoplasma gondii (T. gondii) DNA in blood can help to diagnose the disease in its acute phase; however, it must be considered that hemoglobin, present in blood, can inhibit polymerase activity, making impracticable the detection of DNA in samples. Mice were experimentally infected via oral route with ME49 and BTU2 strains cysts and RH strain tachyzoites; polymerase chain reaction was used to detect T. gondii DNA in mice sera 18, 24, 48, 96, and 192 hours post infection (PI). Toxoplama gondii DNA was detected in only one animal infected with BTU2 strain, genotype III (isolated from a dog with neurological signs) 18 hours PI. The agent's DNA was not detected in any sample of the other experimental groups. New studies must be carried out to...

Brandão,Laila Natasha Santos; Rosa,Janaina Marcela Assunção; Kramer,Beatris; Sousa,Alessandra Tammy Hayakawa Ito de; Trevisol,Iara Maria; Silva,Virginia Santiago; Nakazato,Luciano; Dutra,Valéria.
ABSTRACT: Wild boars (Sus scrofa) have become an important invasive species in all Brazilian regions. Increase in their population causes damage to rural properties, as they invade and destroy crops. To protect their crops and farm animals, producers hunt wild boars and often consume the product without any sanitary control, becoming exposed to various types of pathogens, including Toxoplasma gondii. Sanitary evaluations of these animals are scarce, especially in relation to the protozoan T. gondii. This study aimed to evaluate the occurrence of this pathogen in wild boars in Brazil. We analyzed 122 blood samples from wild boars (blood clots and serum), collected between 2014 and 2016 in five Brazilian states, using polymerase chain reaction (PCR) and...

Importance of high IgG anti-Toxoplasma gondii titers and PCR detection of T. gondii DNA in peripheral blood samples for the diagnosis of AIDS-related cerebral toxoplasmosis: a case-control study BJID
Vidal,José E; Diaz,Adrián Vladimir Hernández; Oliveira,Augusto César Penalva de; Dauar,Rafi Felicio; Colombo,Fabio Antonio; Chioccola,Vera Lucia Pereira.
BACKGROUND: Cerebral toxoplasmosis (CT) continues to cause significant morbidity and mortality in human immunodeficiency virus (HIV)-infected patients in Brazil. In clinical practice, the initial diagnosis is usually presumptive and alternative diagnosis tools are necessary. Our objective was to evaluate whether the detection of high titers of IgG anti-Toxoplasma gondii and T. gondii DNA in blood samples are associated with the diagnosis of CT. METHODS: In this case-control study we included 192 patients with HIV-1 infection: 64 patients with presumptive CT (cases) and 128 patients with other diseases (controls). Blood samples to perform indirect immunofluorescense reaction (IFI) to detect anti-T. gondii IgG antibodies and polymerase chain reaction (PCR)...

Inorganic nanoparticles kill Toxoplasma gondii via changes in redox status and mitochondrial membrane potential OAK
Adeyemi, Oluyomi Stephen; Murata, Yuho; Sugi, Tatsuki; Kato, Kentaro.
This study evaluated the anti-Toxoplasma gondii potential of gold, silver, and platinum nanoparticles (NPs). Inorganic NPs (0.01-1,000 mu g/mL) were screened for antiparasitic activity. The NPs caused >90% inhibition of T. gondii growth with EC50 values of <= 7, <= 1, and <= 100 mu g/mL for gold, silver, and platinum NPs, respectively. The NPs showed no host cell cytotoxicity at the effective anti-T. gondii concentrations; the estimated selectivity index revealed a >= 20-fold activity toward the parasite versus the host cell. The anti-T. gondii activity of the NPs, which may be linked to redox signaling, affected the parasite mitochondrial membrane potential and parasite invasion, replication, recovery, and infectivity potential. Our results...

Measles, rubella, mumps and Toxoplasma gondii antibodies in saliva of vaccinated students of schools and universities in São Paulo City, Brazil BJID
Sampaio,Barbara Carvalho Fialho; Rodrigues,Jaqueline Polizeli; Meireles,Luciana Regina; Andrade Junior,Heitor Franco de.
ABSTRACT Introduction: Vaccines are well-established public health interventions with major impacton the prevalence of infectious diseases, but outbreaks are occurring frequently due to pri-mary and secondary failures, despite high coverage. Surveillance of efficacy and duration ofinduced immunity is a difficult task as it requires invasive blood sampling in children andteenagers. Saliva can be an acceptable alternative source of IgG to assess vaccine efficacyand toxoplasmosis incidence. We investigated IgG response for measles, mumps, rubella, and T. gondii in saliva samples of vaccinated young people. Methods: Saliva was collected from 249 public schools students from São Paulo, Brazil, aged7 to 13 years old, during an interactive exhibition on hygiene....

Toxoplasma gondii infects humans through the gastrointestinal tract (GIT), which elicits humoral immune response with specific antibodies. The expression of the ABO blood group glycoconjugates also occurs in this same system and may influence the human susceptibility of infection by T. gondii. The aim of the present study was to investigate the association between ABO blood group phenotypes and the presence of anti-T. gondii antibodies. Data - including age, results of serology tests for T. gondii infection and ABO blood group phenotypes - were assembled from the medical records of 1,006 pregnant women attended in the Base Hospital of the Medical School of São José do Rio Preto, Brazil, between 2001 and 2004. The chi-square test was used to compare the...

Postnatal acquired toxoplasmosis patients in an infectious diseases reference center BJID
Silva,Cassius Schnell Palhano; Neves,Elizabeth de Souza; Benchimol,Eliezer Israel; Moraes,Danielle Ribeiro de.
Infection caused by Toxoplasma gondii, toxoplasmosis, is one of the most frequent zoonoses in the world; it normally affects both genders equally. Humans are one of several possible intermediate hosts, and the disease is oligosymptomatic in most cases. Vertical transmission is an important cause of fetal malformation and sequels in newborns. Approximately 10% of postnatal cases present multiple manifestations, ranging from low fever and mild lymphadenopathy to severe encephalitis. In moderate cases, lesions such as retinochoroiditis may emerge during acute infection or even years later. We analyzed 313 cases of toxoplasmosis from 1992 to 2004, including 261 acute cases. Most patients were women (68.1%), and 39% of these were pregnant. Among acute infection...
